Tiivistelmä
Entisten jätehuollon käytäntöjen seurauksena kaatopaikoille on kertynyt suuria määriä jätettä vuosien saatossa. Tästä jätteestä suuri osa on orgaanista ainesta, joka hajoaa anaerobisissa oloissa biologisesti muodostaen metaania sisältävää kaatopaikkakaasua. Kaatopaikkakaasun kerääminen mahdollistaa metaanin sisältämän energian hyötykäyttämisen, jolloin myös jätteen sisältämä energia saadaan osittain hyödynnettyä. Metaanin muodostamisen tehostamiseksi kaatopaikoista on rakennettu bioreaktoreita, joissa orgaanisen aineksen biologiselle hajoamiselle pyritään muodostamaan mahdollisimman suotuisat olosuhteet. Kaikki orgaaninen aines ei kuitenkaan hajoa anaerobisissa oloissa. Yksi tällainen materiaali on ligniini, joka esiintyy kaatopaikoilla osana lignoselluloosaa. On arvioitu, että ligniini muodostaisi jopa 10–35 % ikääntyvien kaatopaikkabioreaktoreiden jätemateriaalista.
Tässä työssä arvioitiin ikääntyvään kaatopaikkabioreaktoriin kertyvän ligniinin hajottamista entsyymien avulla. Ligniinin depolymerisaatiota katalysoivia entsyymeitä ovat peroksidaasit ja lakkaasi. Peroksidaasien soveltumista arvioitiin kirjallisuuden perusteella ja lakkaasin soveltumista laboratoriokokeella. Lisäksi arvioitiin ikääntyvän kaatopaikan soveltumista entsymaattiseen käsittelyyn sen olosuhteiden osalta yleisesti sekä erityisesti Ämmässuon vanhan kaatopaikan osalta.
Laboratoriokokeen perusteella lakkaasi nopeutti kaatopaikkakaasun ja metaanin muodostumista jätetäyttöön tehdyn ilmastuksen ja entsyymilisäyksen jälkeen verrattuna pelkkään ilmastukseen. Anaerobiseen hajoamiseen verrattuna lakkaasikäsittely tuotti saman verran metaania, mutta lyhyemmässä ajassa. Ilmastaminen kasvatti orgaanisen aineksen hajoamisnopeutta anaerobiseen hajoamiseen verrattuna.
Ämmässuon vanhan kaatopaikan arvioitiin elinkaarensa puolesta olevan niin sanotussa ilman tunkeutumisvaiheessa, jossa biologisesti saatavilla olevan hiilen puutteen vuoksi anaerobinen hajoaminen ja tätä kautta kaatopaikkakaasun muodostus heikentyy voimakkaasti. Elinkaaren vaiheen perusteella kaatopaikka soveltuisi entsymaattiseen käsittelyyn, mutta epävarmuudet kaatopaikan sisältämän jätteen koostumuksen sekä erityisesti suotoveden kulkeutumisen osalta aiheuttavat merkittävän riskin entsymaattisen käsittelyn onnistumiselle.
Vanha kaatopaikan mahdolliseen entsymaattiseen käsittelyyn on tämän tutkimuksen perusteella kaksi vaihtoehtoista ratkaisua, käsittely ilmastamalla ja lakkaasilisäyksellä tai käsittely peroksidaasilisäyksellä. Ilmastamalla ja lakkaasilisäyksellä jätetäytön olosuhteiden on oltava aerobiset, muuten lakkaasi ei katalysoi ligniinin hajoamista. Metaania ei muodostu käsittelyn aikana vaan hajoamisen lopputuotteena syntyy hiilidioksidia. Onnistuneella käsittelyllä saavutetaan jätteen tehokkaampi hajoaminen ja täten nopeampi stabiloituminen. Mikäli käsittelyn jälkeen täyttöön muodostetaan anaerobiset olosuhteet, on mahdollista, että metaanin muodostuminen jatkuu tehokkaammin ja osa lignoselluloosasta, joka ei jatkuvissa anaerobisissa olosuhteissa hajoaisi, saataisiin näin hyödynnettyä energiana. Peroksidaasilisäyksessä olosuhteet voidaan puolestaan pitää anaerobisina, kun peroksidaasin mukana lisätään vetyperoksidia. Anaerobisten olosuhteiden säilyttäminen mahdollistaa metaanin muodostumisen jatkumisen käsittelyn ajan. Onnistuneen käsittelyn seurauksena jätteen hajoaminen tehostuu peroksidaasin mahdollistaessa ligniinin hajoamisen, täten nopeuttaen jätteen stabiloitumista.
As a result of past waste management practices, large amounts of waste have accumulated in landfills over the years. Much of this waste is organic matter that biodegrades under anaerobic conditions to form methane-containing landfillgas. The collection of the landfillgas makes it possible to utilize the energy potential of the methane, and thus the energy potential of the waste. In order to increase the formation of the methane in landfills, landfill bioreactors have been constructed to optimize the conditions for the biodegradation of the organic matter. However, not all organic matter degrades under anaerobic conditions. One such material is lignin, which is present in landfills as part of lignocellulose. It has been estimated that lignin would make up to 10-35% of the waste material in an aging landfill bioreactor.
In this study, the degradation of lignin, accumulating in the aging landfill bioreactor, using enzymes was evaluated. Enzymes that catalyse the depolymerization of lignin include peroxidases and laccase. The suitability of peroxidases was evaluated based on the literature and the suitability of laccase in a flow-through reactor experiment. In addition, the suitability of the aging landfill for enzymatic treatment was assessed in terms of its conditions. Based on the gathered information the enzymatic treatment of the old landfill in Ämmässuo was evaluated.
The laboratory experiment indicates that laccase enhanced the formation of landfillgas and methane after aeration and enzyme addition compared to aeration alone. When compared to anaerobic degradation, the laccase treatment produced the same amount of methane, but in a shorter time frame. Aeration increased the rate of decomposition of organic matter compared to anaerobic decomposition.
Regarding to the life cycle, the old landfill in Ämmässuo was estimated to be in the so-called air intrusion phase, where due to the lack of bioavailable carbon, anaerobic degradation and thus landfillgas formation is severely impaired. Based on the assessment of the life cycle, the landfill would be suitable for enzymatic treatment, but uncertainties regarding the composition of the waste and in particular the leachate flow inside the landfill, pose a significant risk to the success of the treatment.
Based on this study, there are two alternative solutions for possible enzyme treatment of the old landfill in Ämmässuo. Treatment by aeration with laccase addition or treatment with peroxidase addition. Treatment by aeration with laccase addition requires aerobic conditions inside the landfill, otherwise the laccase will not catalyze the degradation of lignin. Methane is not formed during the treatment and carbon dioxide is produced as a final product. Successful treatment would achieve more efficient degradation of the waste and thus faster stabilization. If, after the treatment, anaerobic conditions are formed, it is possible that the formation of methane could continue and part of the lignocellulose which would have not degraded under continuous anaerobic conditions could thus be utilized as energy. In the case of peroxidase addition, the conditions can remain anaerobic when hydrogen peroxide is added with the peroxidase. Maintaining anaerobic conditions allows methane formation to continue during the treatment. As a result of the successful treatment, the degradation of the waste is enhanced due the peroxidase allowing the degradation of the lignin, thus improving the methane generation and accelerating the stabilization of the waste.
Tässä työssä arvioitiin ikääntyvään kaatopaikkabioreaktoriin kertyvän ligniinin hajottamista entsyymien avulla. Ligniinin depolymerisaatiota katalysoivia entsyymeitä ovat peroksidaasit ja lakkaasi. Peroksidaasien soveltumista arvioitiin kirjallisuuden perusteella ja lakkaasin soveltumista laboratoriokokeella. Lisäksi arvioitiin ikääntyvän kaatopaikan soveltumista entsymaattiseen käsittelyyn sen olosuhteiden osalta yleisesti sekä erityisesti Ämmässuon vanhan kaatopaikan osalta.
Laboratoriokokeen perusteella lakkaasi nopeutti kaatopaikkakaasun ja metaanin muodostumista jätetäyttöön tehdyn ilmastuksen ja entsyymilisäyksen jälkeen verrattuna pelkkään ilmastukseen. Anaerobiseen hajoamiseen verrattuna lakkaasikäsittely tuotti saman verran metaania, mutta lyhyemmässä ajassa. Ilmastaminen kasvatti orgaanisen aineksen hajoamisnopeutta anaerobiseen hajoamiseen verrattuna.
Ämmässuon vanhan kaatopaikan arvioitiin elinkaarensa puolesta olevan niin sanotussa ilman tunkeutumisvaiheessa, jossa biologisesti saatavilla olevan hiilen puutteen vuoksi anaerobinen hajoaminen ja tätä kautta kaatopaikkakaasun muodostus heikentyy voimakkaasti. Elinkaaren vaiheen perusteella kaatopaikka soveltuisi entsymaattiseen käsittelyyn, mutta epävarmuudet kaatopaikan sisältämän jätteen koostumuksen sekä erityisesti suotoveden kulkeutumisen osalta aiheuttavat merkittävän riskin entsymaattisen käsittelyn onnistumiselle.
Vanha kaatopaikan mahdolliseen entsymaattiseen käsittelyyn on tämän tutkimuksen perusteella kaksi vaihtoehtoista ratkaisua, käsittely ilmastamalla ja lakkaasilisäyksellä tai käsittely peroksidaasilisäyksellä. Ilmastamalla ja lakkaasilisäyksellä jätetäytön olosuhteiden on oltava aerobiset, muuten lakkaasi ei katalysoi ligniinin hajoamista. Metaania ei muodostu käsittelyn aikana vaan hajoamisen lopputuotteena syntyy hiilidioksidia. Onnistuneella käsittelyllä saavutetaan jätteen tehokkaampi hajoaminen ja täten nopeampi stabiloituminen. Mikäli käsittelyn jälkeen täyttöön muodostetaan anaerobiset olosuhteet, on mahdollista, että metaanin muodostuminen jatkuu tehokkaammin ja osa lignoselluloosasta, joka ei jatkuvissa anaerobisissa olosuhteissa hajoaisi, saataisiin näin hyödynnettyä energiana. Peroksidaasilisäyksessä olosuhteet voidaan puolestaan pitää anaerobisina, kun peroksidaasin mukana lisätään vetyperoksidia. Anaerobisten olosuhteiden säilyttäminen mahdollistaa metaanin muodostumisen jatkumisen käsittelyn ajan. Onnistuneen käsittelyn seurauksena jätteen hajoaminen tehostuu peroksidaasin mahdollistaessa ligniinin hajoamisen, täten nopeuttaen jätteen stabiloitumista.
